An exciting opportunity exists for a Research Technician to join the research team in the MacKinnon Lab of the Howard Hughes Medical Institute at the Rockefeller University. The Research Technician will perform a broad range of experiments in the areas of biochemistry, molecular and cell biology to contribute to several research projects. This laboratory is working on the identification, expression, purification, crystallization and functional characterization of membrane ion channel proteins. Targets are identified, their genes cloned or synthesized, and protein expressed and purified in quantities sufficient for biochemical and electrophysiological characterization, as well as crystallization for elucidation of structure by X-ray diffraction. Principal Responsibilities: Designing oligonucleotides for sequencing and mutagenesisCloning natural and synthetic genes into vectorsPCR, cloning, site-directed mutagenesis and DNA sequence analysisTransfection and testing for expression in a variety of biological systems including prokaryotic, yeast, mammalian and baculovirusMaintenance of plasmid stocks, inventory, ordering of molecular biology reagents and organizing sequence recordsDevelop, validate and document general molecular biology protocols to be use by the laboratory as a wholeWork individually and as an effective member of a team on particular projects

' May review run binders and notebooks and other project-related documentation at the direction of the Project Leader (excludes validation reports).
' Assists in maintaining adequate supplies.

Survey Senior Research Associate
The Senior Research Associate will work alongside Senior Analysts and will have the following primary duties:
o Manage online surveys throughout the entire project life cycle. Tasks include:
o Edit survey questionnaires to fit respondent friendly formats and programming formats.
o Work with programmers to co-ordinate and test programming of questionnaires into online survey formats
o Research, identify, and recruit appropriate respondents needed to meet quotas for survey recruiting.
o Manage relationships with survey respondents and answer all technical and non-technical questions pertaining to the survey.
o Analysis of large quantities of data from surveys into client-deliverable formats using SPSS, Microsoft Excel, Word, and PowerPoint,

The technician will participate in our NIH-funded Structural Genomics program. They will have the opportunity to train on and utilize state-of-the art equipment.
Responsibilities of Research Technician B Includes:
Duties would involve robotic protein crystallization and optimization and crystal freezing.
The technician will visually evaluate crystal quality and protein purity and evaluate the results of thermal denaturation experiments.
Additional duties would involve simple protein chemistry techniques, such as centrifugation and melting temperature determinations.
The technician will work closely with the research team to adjust solutions as necessary.
They will also be required to document procedures and provide short contributions for quarterly reports to the NIH describing the crystallization and protein chemistry protocols.

The analyst will be responsible for operation of the GC and associated purge and trap concentrator as well as FID, Hall Cell and PID detectors.
Must be familiar with the operation and calibration techniques of gas chromatography.
